MiCA-Inv: Microfluidic Cell Array for Cancer Invasion

A breakthrough microfluidic perfusion plate that enables dynamic tracking of cancer cell invasion through 3D matrix. The MiCA-Invasion array contains 24 independent micro-chambers that mimic the micro-environment of cancer invasion in vivo. The chambers are loaded by the user with an extracellular matrix (ECM) and cells are cultured with continuous perfusion. Perform multiple 3D invasion experiments at a fraction of the time and cost of alternate methods. Formatted to a standard 96 well plate with a #1.5 thickness glass bottom, enabling high quality microscopy. All products backed by our 100% Customer Satisfaction Guarantee.

Key Features:

• 24 independent perfusion units
• Bio-mimetic invasion chamber design
• Use only 5 ul of gel per micro-chamber
• Continuous perfusion 3D culture
• Pump-free culture in a standard incubator
• #1.5 glass bottom for optimal imaging quality

Description:

The MiCA-Inv (M24I) microfluidic plate enables dynamic tracking of cancer invasion through 3D gel in a 96 well format. The invasion chamber is designed to mimic the in vivo microenvironment, with cells embedded in 3D ECM and a set of 8 micron pores separating the cells from continuous flow of media. Cells and gels are loaded by the user into one side of the barrier, while flow of media past the perfusion barriers allows nutrient exchange for long term culture. Solution flows from the inlet well past the cell chamber and collects in the outlet well. The flow is gravity driven and requires no external pumps, allowing the plates to be operated in a standard incubator.

M24I Plate Layout
(a) The M24I microfluidic plate contains 24 perfusion units on a 96 well plate. (b) Each unit has a flow inlet, a cell/gel inlet, an invasion chamber, and an outlet. (c) The invasion chamber is designed for monitoring cells crossing a gel filled invasion barrier (8 um pores) towards a continuous flow channel.

The 50 micron height chamber with thin glass bottom allows high quality live cell imaging with an inverted microscope. Cells can be observed moving through the barrier or screened at endpoints by counting the numbers on either side of the barrier. Compared with current methods for cancer invasion assay, the M24I plate offers higher quality biological information while saving time and materials.

M24I Invasion
Cancer invasion of HT-1080 cells in Matrigel at t=0 (a) and t=24 hours (b). Images taken with a 4X objective. The cells cross the 8 um gel filled invasion barrier (c) in response to stimulus. Close-up image taken with 20X lens.
