Operation

This page describes the basic operation of the 3D:M microfluidic plate. For additional detail or questions, please call or email info@cellasic.com
 

0. The 3D:M plate is shipped pre-primed with PBS and sealed in a sterile pack.

1. In a sterile environment, remove the foil seal.

2. Aspirate all wells of unit to be used. Each unit has 4 wells, as shown. Wells 1, 3, and 4 should be fully aspirated, including the cutout at the bottom of the well (blue outlines).

3. Pipette 5ul of your cell or cell/gel solution into the Cell/Gel Inlet well. Make sure to dispense into the hole at the bottom of the well (blue outline).

The cell/gel solution will load the microchamber via capillary flow. Click here to see a movie.

4. Add 300ul of culture medium to the Medium Inlet well.

5. Place in a standard cell culture incubator. The 3D:M is designed to perfuse the cells at a rate of 40ul/day as medium flows from the Medium Inlet to the Outlet via gravity. Microfluidic air channels enable free gas diffusion into the culture chamber.

6. For long term culture, refill the Medium Inlet and empty the Oulet every 3 days.
